The multibillion dollar CNS drug market is rapidly growing and bears significant unmet medical needs. Many drugs are currently not available to the brain because they are not able to enter the brain due to the presence of the so called blood-brain barrier (BBB). For instance, the brain is a sanctuary site for metastasis of breast tumors normally curable by therapeutic anti-cancer antibodies that are not able to cross the blood-brain barrier, making this type of therapy overall ineffective for these patients. CNS drug targeting technology may solve this problem.
 
to-BBB developed state-of-the-art proprietary technologies that create safe drug delivery opportunities to the brain. The clinical development of the G-Technology^®, which utilizes the endogenous ligand glutathione to target drug-loaded liposomes to the brain, is to-BBB’s primary focus. This second generation technology enables superior transport of drugs across the blood-brain barrier and a better safety- and pharmacokinetic profile compared to competing technologies. The G-Technology^® is already successfully validated in vitro and in animal models for Brain Tumors, pain and Viral Encephalitis.

The founding fathers of to-BBB were Pieter Gaillard, Alfred Nijkerk, Bert de Boer and Leiden University who set up the company in 2003 as a spin-off from the renowned blood-brain barrier research group of the Leiden/Amsterdam Center for Drug Research from Leiden University in The Netherlands. 

to-BBB's business model is two-fold:                                                                           (1) Value generation through focused internal development of brain-targeted liposomal formulations of off-patent medications;                                                                       (2) Generating license income by partnering with pharma companies to deliver their patented compounds in to-BBB’s targeted liposomes to the brain.
